Comparative cytogenetics of Hoplias malabaricus (Pisces, Erythrinidae): a population analysis in adjacent hydrographic basins Genet. Mol. Biol.
Vicari,Marcelo Ricardo; Artoni,Roberto Ferreira; Bertollo,Luiz Antonio Carlos.
The chromosomes of specimens from four Hoplias malabaricus populations from headwaters of adjacent river basins at Ponta Grossa, southern Brazil, were investigated using differential staining techniques (C-banding, AgNO3 and CMA3) and fluorescent in situ hybridization (FISH) with an 18S rDNA probe. The diploid chromosome number in representatives of all four populations was invariably 2n = 42, with karyotypes composed of 12 pairs of metacentrics and 9 pairs of submetacentrics, without heteromorphic sex chromosomes. This kind of karyotype represents cytotype A in regard to cytotypes identified previously in H. malabaricus, exhibiting however, at the same time, some differences in the distribution of constitutive heterochromatin segments and in the locations...

Comparative cytogenetics of three species of Dichotomius (Coleoptera, Scarabaeidae) Genet. Mol. Biol.
Silva,Guilherme Messias da; Bione,Edgar Guimarães; Cabral-de-Mello,Diogo Cavalcanti; Moura,Rita de Cássia de; Simões,Zilá Luz Paulino; Souza,Maria José de.
Meiotic and mitotic chromosomes of Dichotomius nisus, D. semisquamosus and D. sericeus were analyzed after conventional staining, C-banding and silver nitrate staining. In addition, Dichotomius nisus and D. semisquamosus chromosomes were also analyzed after fluorescent in situ hybridization (FISH) with an rDNA probe. The species analyzed had an asymmetrical karyotype with 2n = 18 and meta-submetacentric chromosomes. The sex determination mechanism was of the Xy p type in D. nisus and D. semisquamosus and of the Xyr type in D. sericeus. C-banding revealed the presence of pericentromeric blocks of constitutive heterochromatin (CH) in all the chromosomes of the three species. After silver staining, the nucleolar organizer regions (NORs) were located in...

Comparative karyotype analysis of populations in the Alstroemeria presliana Herbert (Alstroemeriaceae) complex in Chile Genet. Mol. Biol.
Baeza,Carlos; Finot,Víctor L.; Ruiz,Eduardo.
Alstroemeria L., one of the most diverse genera of the Chilean flora and of high floricultural value, is represented by 35 species, most of them distributed between 28–38° S in the Mediterranean zone of Central Chile. There are 24 complex-forming taxa, of which 18 have conservation problems (8 are considered “endangered” and 10 as “vulnerable”). One of these complexes is Alstroemeria presliana Herb. with two subspecies: subsp. presliana and subsp. australis Bayer. Alstroemeria presliana grows in Chile and Argentina: subsp. presliana is distributed from Reserva Nacional Siete Tazas (35°27′ S, Region of Maule) to Antuco, (37°25′ S, Region of Bío-Bío), and is also found in Neuquén, Argentina; subsp. australis is endemic to the Cordillera of Nahuelbuta. Comparative karyotypic analysis in the Alstroemeria hookeri Lodd. (Alstroemeriaceae) complex sensu Bayer (1987) Genet. Mol. Biol.
Baeza,Carlos; Ruiz,Eduardo; Negritto,María.
Alstroemeria L. (Alstroemeriaceae) is an American genus of monocots with two principal distribution centers in Chile and Brazil. In Chile, it is represented by about 32 species, most of them in central Chile, an area known for its high level of endemism. The "complex" Alstroemeria hookeri is endemic to Chile, where it is distributed from the Coquimbo to the Bío-Bío Region. We analyzed the karyotypes of 36 populations of this complex along its natural distribution. Ten metaphases per population were used for chromosome measurements. All analyzed subspecies presented a well defined asymmetric karyotype. The populations of A. hookeri subsp. hookeri collected in the coastal range of the Bío-Bío Region and the populations from the Central Valley of this Region...

Cytogenetic analysis of five species of the subfamily Corydoradinae (Teleostei: Siluriformes: Callichthyidae) Genet. Mol. Biol.
Shimabukuro-Dias,Cristiane Kioko; Oliveira,Claudio; Foresti,Fausto.
In the present study, five callichthyid species belonging to the subfamily Corydoradinae were karyotyped: three species of Aspidoras and two of Corydoras. The three species of Aspidoras had the same diploid number, 2n = 46 chromosomes, similar karyotypic formulae, with most chromosomes metacentric or submetacentric, single interstitial Ag-NORs and C-band positive segments mainly found in the centromeric position. The comparative analysis of cytogenetic data available for the genus Aspidoras and other species of Corydoradinae suggest that several events of centric fusion occurred in the origin of the species of Aspidoras. The two analyzed species of Corydoras showed high diploid numbers, 2n = 74 in C. sodalis and 2n = 90 in C. britskii. While C. sodalis...

Cytogenetic and morphological diversity in populations of Astyanax fasciatus (Teleostei, Characidae) from Brazilian northeastern river basins Genet. Mol. Biol.
Medrado,Aline Souza; Figueiredo,Alba Vivian Amaral; Waldschmidt,Ana Maria; Affonso,Paulo Roberto Antunes de Mello; Carneiro,Paulo Luiz Souza.
In the present work, morphometric and cytogenetic analyses were carried out in populations of the fish Astyanax fasciatus (Characidae) from Contas and Recôncavo Sul River basins (State of Bahia, Brazil), providing new data on the genetic structure of this species along the region. Based on morphologic measurements, we observed that populations from the same hydrographic basin were more similar to each other (Contas and Preto do Costa Rivers), and remarkably divergent from Recôncavo Sul (Mineiro Stream), as indicated by clustering analysis. Cytogenetic data revealed a same diploid number for all populations (2n = 48), but distinct karyotype formulae (8M+24SM+12ST+4A, FN = 92 in Contas River, 8M+24SM+10ST+6A, FN = 90 in Preto do Costa River, and...

Cytogenetic comparison of Podocnemis expansa and Podocnemis unifilis: a case of inversion and duplication involving constitutive heterochromatin Genet. Mol. Biol.
Gunski,Ricardo José; Cunha,Isabel Souza; Degrandi,Tiago Marafiga; Ledesma,Mario; Garnero,Analía Del Valle.
Podocnemis expansa and P. unifilis present 2n = 28 chromosomes, a diploid number similar to those observed in other species of the genus. The aim of this study was to characterize these two species using conventional staining and differential CBG-, GTG and Ag-NOR banding. We analyzed specimens of P. expansa and P. unifilis from the state of Tocantins (Brazil), in which we found a 2n = 28 and karyotypes differing in the morphology of the 13th pair, which was submetacentric in P. expansa and telocentric in P. unifilis. The CBG-banding patterns revealed a heterochromatic block in the short arm of pair 13 of P. expansa and an interstitial one in pair 13 of P. unifilis, suggesting a pericentric inversion. Pair 14 of P. unifilis showed an insterstitial band in...

Cytogenetic data of Partamona peckolti (Hymenoptera, Apidae, Meliponini) by C banding and fluorochrome staining with DA/CMA3 and DA/DAPI Genet. Mol. Biol.
Brito,Rute Magalhães; Caixeiro,Ana Paula de Almeida; Pompolo,Silvia das Graças; Azevedo,Giselle Garcia.
The stingless bees of the Partamona genus have been studied taxonomically, ecologically and behaviourally, but cytogenetic studies are still rare. The objective of this study was to obtain cytogenetic data to contribute to Partamona peckolti species characterization. Heterochromatin was localized in all chromosome pericentromeric regions but some blocks could be visualized on some large chromosomes arms. A large heterozygous DA-CMA3-positive band was observed on one large chromosome arm, but was completely absent when C banding was applied before fluorochrome staining, with only one small positive band being visualized. Sequential DA-CMA3-NOR staining of interphase nuclei provided coincident positive responses. This suggests that DA-CMA3-positive bands of...

Cytogenetic studies in some species of Passiflora L. (Passifloraceae): a review emphasizing Brazilian species BABT
Souza,Margarete Magalhães; Pereira,Telma N. Santana; Vieira,Maria Lúcia Carneiro.
The Passifloraceae is represented by species of tropical and subtropical origin. The Passiflora is the richest genus with approximately 450 species, 200 of them being native to Brazil. Recent karyological studies have reported the basic chromosome number for the Passiflora genus as x = 6, whereas x = 9, x = 10 and x = 12 were established as secondary basic numbers. High rates of fertility occur in most Passiflora species, since both meiotic index and pollen viability are above 90%. Unusual meiotic behavior has been described in some taxa. Unviable pollen were observed in some diploids species. The genome size varies from 1.83 to 5.36 pg, and significant interspecific variance has been observed. Studies using the FISH methodology have shown that there are...

Cytogenetic studies in three species of Lutjanus (Perciformes: Lutjanidae: Lutjaninae) from the Isla Margarita, Venezuela Neotropical Ichthyology
Nirchio,Mauro; Rondón,Rodolfo; Oliveira,Claudio; Ferreira,Irani A.; Martins,Cesar; Pérez,Julio; Sola,Luciana; Rossi,Anna Rita.
In the present study, three species of Lutjaninae, Lutjanus analis, L. griseus and L. synagris, were analyzed by conventional Giemsa staining, C-banding and silver staining, to reveal active Nucleolus Organizer Regions (NORs). Fluorescent in situ hybridization (FISH) was also applied to establish the number and location of the ribosomal gene clusters (18S and 5S rRNA genes). Counts of diploid metaphasic cells revealed a diploid modal chromosome complement composed of 48 acrocentric chromosomes in both L. analis and L. griseus. Two cytotypes were observed in L. synagris: cytotype I, with 2n=48 acrocentric chromosomes, found in 19 specimens, and cytotype II, with 46 acrocentric chromosomes and one large metacentric, found in two specimens. The large...

Cytogenetic studies of Brazilian pediatric myelodysplastic syndrome cases: challenges and difficulties in a large and emerging country BJMBR
Velloso,E.D.R.P.; Chauffaille,M.L.; Peliçario,L.M.; Tanizawa,R.S.S.; Toledo,S.R.C.; Gaiolla,R.D.; Lopes,L.F..
Myelodysplastic syndromes (MDS) and juvenile myelomonocytic leukemia (JMML) are rare hematopoietic stem cell diseases affecting children. Cytogenetics plays an important role in the diagnosis of these diseases. We report here the experience of the Cytogenetic Subcommittee of the Brazilian Cooperative Group on Pediatric Myelodysplastic Syndromes (BCG-MDS-PED). We analyzed 168 cytogenetic studies performed in 23 different cytogenetic centers; 84 of these studies were performed in patients with confirmed MDS (primary MDS, secondary MDS, JMML, and acute myeloid leukemia/MDS+Down syndrome). Clonal abnormalities were found in 36.9% of the MDS cases and cytogenetic studies were important for the detection of constitutional diseases and for differential diagnosis...

Cytogenetic studies of three Lycosidae species from Argentina (Arachnida, Araneae) Genet. Mol. Biol.
Chemisquy,María A.; Gil,Sergio G. Rodríguez; Scioscia,Cristina L.; Mola,Liliana M..
Cytogenetic studies of the family Lycosidae (Arachnida: Araneae) are scarce. Less than 4% of the described species have been analyzed and the male haploid chromosome numbers ranged from 8+X 1 X 2 to 13+X 1 X 2 . Species formerly classified as Lycosa were the most studied ones. Our aim in this work was to perform a comparative analysis of the meiosis in " Lycosa " erythrognatha Lucas, " Lycosa " pampeana Holmberg and Schizocosa malitiosa (Tullgren). We also compared male and female karyotypes and characterized the heterochromatin of " L. " erythrognatha . The males of the three species had 2n = 22, n = 10+X 1 X 2 , all the chromosomes were telocentric and there was generally a single chiasma per bivalent. In " Lycosa " pampeana , which is...

Cytogenetic study in natural hybrids of Callithrix (Callitrichidae: Primates) in the Atlantic forest of the state of Rio de Janeiro, Brazil Iheringia, Sér. Zool.
Nogueira,Denise M.; Ferreira,Ana Maria R.; Goldschmidt,Beatriz; Pissinatti,Alcides; Carelli,Juliane B.; Verona,Carlos E..
In the Atlantic forest of Rio de Janeiro, Callithrix aurita (É. Geoffroy in Humboldt, 1812) is a native species vulnerable to extinction and C. jacchus (Linnaeus, 1758) and C. penicillata (É. Geoffroy, 1812) are invasive species. The major threats to the native species are habitat degradation and hybridization, although there are currently no genetic data about natural hybrids available. Previous studies have revealed that species of the Callithrix genus are extremely homogeneous in their karyotypes with the exceptions of the morphology and size of the Y chromosome and its nucleolar organizer region (NOR) banding pattern. Three male marmosets captured in the wild in Guapimirim municipality, Rio de Janeiro, Brazil, considered as possible hybrids between C....

Cytogenetic study of the giant otter Pteronura brasiliensis Zimmermann 1780 (Carnivora, Mustelidae, Lutrinae) Genet. Mol. Biol.
Franco-de-Sá,Jorge Felipe Oliveira; Rosas,Fernando César Weber; Feldberg,Eliana.
The giant otter, Pteronura brasiliensis Zimmermann 1780 (Carnivora, Mustelidae, Lutrinae), was widely distributed in South America but stable populations are now only found in the Pantanal and Amazon regions and the species is classified as endangered. There is only one recognized species of giant otter, although two subspecies of doubtful value have also been cited in the literature. We present the first karyotype of four captive P. brasiliensis specimens, all of which posses 2n = 38 chromosomes as 14M+8SM+6ST+8A and one pair of sexual chromosomes. An heteromorphic secondary constriction, associated with the nucleolar organizer region (NOR), was seen on the long arms of chromosome pair 17. The C-banding technique revealed heterochromatin in the...

Cytogenetical and morphological features reveal significant differences among Venezuelan and Brazilian samples of Mugil curema (Teleostei: Mugilidae) Neotropical Ichthyology
Nirchio,Mauro; Cipriano,Roger; Cestari,Margarete; Fenocchio,Alberto.
Karyotype of M. curema from the Gulf of Mexico and Brazil have been reported as possessing chromosome complement with 2n=28 and FN=48, whereas specimens from Venezuela has been reported as possessing a diploid number 2n=24 and a conserved FN (48). Although at first sight this variation suggests the presence of a chromosomal intraspecific (interpopulational) variability, the possibility that we are dealing with two different species was examined. This work revisit the karyotypes of M. curema from Venezuela and Brazil, including new data on C-banding, and NOR localization, and compares morphologic characteristics of samples from both localities. Thus, besides diploid number, the constitutive heterochromatin distribution and NORs location, mark other...

Cytogenetics and Molecular Genetic Analysis of Chimerism in Marmosets (Callithrix: Primates) Anais da ABC (AABC)
SILVA,MONIQUE O.M.; ARMADA,JORGE LUÍS A. DE; VERONA,CARLOS EDUARDO S.; HELIODORO,GABRIELA; NOGUEIRA,DENISE M..
ABSTRACT The birth of fraternal twins is a characteristic frequently observed in callitrichids. Cytogenetic studies have demonstrated hematopoietic chimerism in marmosets with the occurrence of two cell lines 2n=46,XX/46,XY in females and males co-twins, without phenotypic changes. Amplification by PCR have also been used to verify the presence of the SRY gene in female chimaeras. Our aim was to verify the occurrence of chimerism in Callithrix sp. individuals considered as hybrids according to their intermediate phenotypes between C. jacchus and C. penicillata. Blood samples from 37 Callithrix sp. individuals were collected. Hematopoietic chimerism 2n=46,XX/46,XY was detected by cytogenetic analysis in five individuals, three males and two females. A...

Cytogenetics of coffee Braz. J. Plant Physiol.
Pinto-Maglio,Cecília A. F..
The genus Coffea L. has around 100 native species distributed in tropical and subtropical areas in Africa, and the most important economic species are C. arabica and C. canephora. C. arabica is exceptional in the genus since it is the only species so far analyzed which is self-compatible, and a natural polyploid with 2n=4x=44 chromosomes; it is considered to be a segmental allopolyploid because it presents a disomic inheritance and a regular meiotic behavior. All other species in the genus are self-incompatible diploids with 2n=2x=22. Cytogenetic studies in Coffea, undertaken since 1912, have followed various phases: initial studies were limited only to establishing chromosome counts. Subsequent studies characterized the karyotypes of various species using...
